This Fungi/Yeast Genomic DNA Isolation 96-Well Kit offers a fast, reliable, and simple method for high-throughput isolation of DNA from viable yeast cells, fungal spores or mycelium, and Gram-positive bacteria. Genomic DNA is efficiently extracted through a combination of heat treatment, detergents, and bead-based homogenization. The resulting DNA is of high quality and fully compatible with downstream applications such as real-time PCR and sequencing. An optional lyticase treatment is included to enhance DNA yield for certain fungal and yeast species.

Purification Technology

Purification is based on 96-well column chromatography, which selectively isolates DNA from other cellular components, such as proteins, without the use of phenol or chloroform. The procedure can be performed using either a vacuum manifold or centrifugation.

The process begins by adding lysis buffer to pelleted cells, fungi, yeast, or Gram-positive bacteria, followed by transfer to bead tubes for homogenization. Optional lyticase treatment can be applied at this stage. Samples are then incubated at 65°C for 10 minutes, centrifuged, and the supernatant is collected. An equal volume of 96–100% ethanol and binding solution is added to the lysate, which is then applied to a 96-well plate that selectively binds DNA. Bound DNA is washed with the provided wash solution and subsequently eluted using the elution buffer.

The purified total DNA is free of inhibitors and suitable for a wide range of downstream applications, including PCR and sequencing.
