The Phage DNA Isolation Kit provides a fast and efficient method for the isolation and purification of total DNA from bacteriophages propagated in liquid bacterial cultures. The procedure avoids phenol, chloroform, and cesium chloride, offering a safe and reliable alternative for phage DNA extraction.

Designed for small volumes of phage supernatant (as little as 1 mL), the spin column workflow can be completed in under 45 minutes. The purified DNA is of high integrity and suitable for a wide range of molecular biology applications, including Southern blotting, restriction fragment length polymorphism (RFLP), sequencing, cloning, and real-time PCR.

Purification Technology:

Purification is based on spin column chromatography, which selectively binds nucleic acids while removing proteins and contaminants. The workflow includes:

  1. Starting from clarified phage supernatant (free of bacterial debris)
  2. Lysis of phage particles using heat and chemical treatment
  3. Addition of isopropanol and loading of lysate onto a spin column
